Taxus, a genus of slow-growing evergreen shrubs known as yews, produces unique diterpenes called taxanes, found only in Taxus and Austrotaxus spicata. Traditionally, yew extracts were used in folk medicine for various treatments. In the 1950s, the National Cancer Institute began screening plant extracts for tumor models. In 1964, Wall and Wani isolated taxol (paclitaxel) from yew bark, with its full clinical potential recognized in 1974. Taxol revolutionized cancer treatment by promoting irreversible tubulin assembly into microtubules. Scarcity led to extensive synthetic efforts, culminating in total syntheses in 1994 and solving supply issues via semi-synthesis, but production remains challenging. However, the bioactivity of complex taxanes, or cyclotaxanes, is significantly underexplored despite their potential to reverse multidrug resistance and enhance anti-cancer effects. Certain complex taxanes have shown remarkable multidrug-resistance-reversing activity and enhanced the anti-cancer therapeutic effect by increasing the cellular accumulation of vincristine (VCR) in P388/VCR-bearing mice and tumor cells. However, further research into complex taxanes is hindered by low natural availability, non-sustainable resources, and the lack of efficient synthetic routes. Here we will discuss the story of our synthesis of complex taxane family natural product.
