Darobactin and Dynobactin are recently discovered RiPP (ribosomaly synthesized, post-translationally modified peptide) natural products that show promising potent antibacterial properties against Gram-negative pathogens in combination as well as low toxicity against intestinal gut bacteria and human cell lines. Adding further intrigue and promise to this class of antibiotics is it’s apparent ability to evade bacterial resistance through mutation. Both natural products demonstrated a novel mechanism of action targeting BamA, a bacterial insertase within the β-barrel assembly machinery (BAM) complex that facilitates folding and insertion of outer membrane proteins in Gram-negative bacteria. Testimony to their nature as RiPP natural products, Darobactin and Dynobactin exhibit a number of unusual structural features, comprising b-branched amino acids with additional C-C and C-N bond connectivities resulting in a strained system of fused (Darobactin) or linked (Dynobactin) macrocycles. Solutions towards these synthetic challenges will be discussed, leading up to the successful total synthesis of Darobactin and Dynobactin.
